چکیده

Cilia are evolutionarily highly conserved organelles with important functions in many organs. The extracellular component of the cilium protruding from plasma membrane comprises an axoneme composed microtubule doublets, arranged a 9 + 0 conformation primary cilia or 2 motile cilia. These microtubules facilitate transport intraflagellar cargoes along axoneme. They also provide structural stability to cilium, which may play role sensory cilia, where signals received movement fluid. Post-translational modification is well-studied phenomenon, and acetylation on lysine 40 (K40) alpha tubulin prominent It believed that this contributes stabilization Two classes enzymes, histone acetyltransferases deacetylases, mediate regulation acetylation. Here we use genetic approach, immunocytochemistry behavioral tests investigate function deacetylases zebrafish model. By mutating three deacetylase genes ( Sirt2 , Hdac6 Hdac10 ), identify unforeseen for As expected, mutation these leads increased cytoplasmic tubulin, however, surprisingly it caused decreased developing eye, ear, brain kidney. ear eye showed elevated levels mono-glycylated suggesting compensatory mechanism. changes did not affect length morphology functional defects balance was observed, level cilium.

Furry promotes acetylation of microtubules in the mitotic spindle by inhibition of SIRT2 tubulin deacetylase.

The structure and function of microtubules (MTs) are regulated by post-translational modifications of tubulin subunits, such as acetylation of the Lys40 residue of α-tubulin. Regulation of the organization and dynamics of MTs is essential for the precise formation of the mitotic spindle. Spindle MTs are highly acetylated, but the mechanism regulating this acetylation is largely unknown. Furry (...

Acetylation, deacetylation and acyltransfer

N-Substituted aromatic compounds can be metabolized in most species to N-acetylated derivatives that are themselves subject to further enzymatic transformations, including hydrolysis and N,O-acyltransfer. These proceses can either potentiate or ameliorate the biological responses to these N-substituted derivatives. Decreasing the levels of metabolites, such as arylhydroxylamines may, in some sy...

HDAC6 and SIRT2 regulate the acetylation state and oncogenic activity of mutant K-RAS.

UNLABELLED Activating point mutations in K-RAS are extremely common in cancers of the lung, colon, and pancreas and are highly predictive of poor therapeutic response. One potential strategy for overcoming the deleterious effects of mutant K-RAS is to alter its posttranslational modification. Although therapies targeting farnesylation have been explored, and have ultimately failed, the therapeu...

Myc-Nick: A Cytoplasmic Cleavage Product of Myc that Promotes α-Tubulin Acetylation and Cell Differentiation

The Myc oncoprotein family comprises transcription factors that control multiple cellular functions and are widely involved in oncogenesis. Here we report the identification of Myc-nick, a cytoplasmic form of Myc generated by calpain-dependent proteolysis at lysine 298 of full-length Myc. Myc-nick retains conserved Myc box regions but lacks nuclear localization signals and the bHLHZ domain esse...
